Sekuentziazio masiboaren erabilera izaki eukarioten garapena erregulatzen duten geneak identifikatu eta karakterizatzeko

Abstract

Garapen zelularra harizpi itxurako onddoetan zelula ez-espezializatuetan abiatzen da erregulatzaile goiztiarren bitartez, FlbB transkripzio faktorea (TF) barne. Bere eraginpean dauden beste geneen funtzioak identifikatzeko, ΔflbB mutante eta andui basatiaren (WT) transkriptomak konparatu dira RNA-seq teknikaren bitartez. TF honen funtzio erregulatzaile berriak aurkitu dira. Batetik, FlbBren ausentziak metabolito antibakteriano baten ekoizpenerako beharrezkoak diren geneen espresioa handitzen du. Bestalde, FlbBren bitartez erregulatzen den ustezko TF bat ere identifikatu da, UrdA, garapen mota ezberdinen erregulatzailea dena. Emaitzek FlbBren gabeziak garapen mekanismoak eta prozesu metabolikoak desorekatzen dituela erakusten dute.

Development in filamentus fungi is induced in non-specialized cells by a set of early regulators including the transcription factor FlbB. To identify genes under the transcriptional control of FlbB and determine their role, we compared ΔflbB and wild-type transcriptomes through the RNA-seq technique. We found additional regulatory roles for this TF. On one hand, the absence of FlbB increases the expression of a metabolic cluster that synthesizes an antibacterial metabolite. On the other hand, FlbB mediates the transcriptional control of urdA, coding for a putative TF which regulates the balance between the two developmental cycles of filamentous fungi. Taken together, our results open a new holistic outlook to the study of regulatory networks during development.
