Abstract
1 motako diabetesarekin (DM1) asoziatutako polimorfismo ugari RNA luze ez kodetzaileetan (lncRNA) kokatuta daude. Lan honen helburua DM1-rekin asoziatutako lnc13 eta lncBACH2 deritzen lncRNAen karakterizazio funtzionala gauzatzea izan da. Gure emaitzek lnc13-k β-zelulen hantura prozesuan parte hartzen duela baieztatzen dute. Izan ere, lnc13-ak STAT1 eta PCBP2 molekulen arteko elkarrekintza baimentzen du, hantura sustatzen duen STAT1 mRNAren egonkortasuna emendatzen duelarik. Bestalde, lncBACH2–k BACH2 genearen adierazpena erregulatzen duela eta aldi berean hanturak eragindako β-zelulen apoptosian eragina duela ikusi dugu. Gure lanak gaixotasunarekin lotura duten lncRNAk β-zelulen funtzionamenduan eragina dutela erakusten du, eta hortaz, lncRNAk DM1ren patogenesian berebiziko garrantzia eduki dezaketela iradokitzen du.
Several polymorphisms associated with type 1 diabetes (T1D) are located in long non-coding RNAs (lncRNAs). The aim of the present work is to characterize the functional impact of two lncRNAs; lnc13 and lncBACH2. lnc13 is implicated in β-cell inflammation process. Indeed, lnc13 allows the interaction between STAT1 and PCBP2 molecules, inducing the stability of the pro-inflammatory STAT1 mRNA. On the other hand, lncBACH2 controls BACH2 gene expression which is directly related to β-cell apoptosis. Our work shows that T1D-associated lncRNAs are implicated in pancreatic β-cell function, and thus, they may play a crucial role in the pathogenesis of the disease.
