Abstract
Baltsa lipidikoaren kontzeptua proposatu zenetik 20 urte baino gehiago igaro diren arren, nanoegitura horien existentziak oraindik ere eztabaidagai izaten jarraitzen du, ez baitago egitura horien nanoantolaketa ikertzeko zelula bizietan erabili daitekeen metodo ez-inbaditzailerik. Artikulu honetan kolesterol zunda bioortogonal bat garatu da (chol-N3), zeina bereizmen handiko mikroskopiaz baliatuta, 50 nm baino gutxiagoko lipido-baltsak zelulen mintz plasmatikoan zuzenean ikusi eta karakterizatzeko metodo baliagarria den. Zunda honek kolesterol endogenoa ordezkatzen du mintz sintetiko eta zelularretan, asaldurarik gabe.
Although more than 20 years have passed since the concept of lipid raft was proposed, the existence of these nanostructures is still under discussion, as there are no noninvasive methods that can be used in living cells to investigate the nanoorganization of these structures. In this article, a bioorthogonal cholesterol probe (chol-N3) has been developed which, combined with live-cell super-resolution microscopy, is a powerful method for direct observation and characterization of <50 nm lipid rafts in the plasma membrane of living cells. This probe mimics endogenous cholesterol in synthetic and cellular membranes without perturbation.
