Abstract
Irudi digitalen analisian (DIA) oinarritutako metodo bat garatu da pH-a eta fosfato kontzentrazioa kuantifikatzeko kolirio laginetan. Analisiak 96 putzuko mikroplakak eta smartphone batekin lortutako irudiak erabiliz egin dira. Fosfatoen determinazioa molibdeno urdina erreakzioan sortzen den konplexu urdinaren kolorea RGB kolore-sistemako R kanalarekin erlazionatuz egin da. pH-aren determinazioa horiaren eta urdinaren arteko kolore aldaketa ematen duen bromotimol urdina adierazlearekin egin da, PLS eredu baten bidez irudiaren histograma eta pH balioa erlazionatuz. Lortutako emaitzak erreferentziazko metodoak erabiliz balidatu dira, kromatografia ionikoa fosfatoentzat eta potentziometria pH-arentzat.
A method based on digital image analysis (DIA) is described for the quantification of pH and phosphate concentration in eye drops. Analysis were carried out using 96-well microplates and images obtained with a smartphone. The determination of phosphate was made by relating the color of the blue complex formed in the reaction to the R channel of the RGB color system. The pH was measured using bromothymol blue indicator, which gives a color change between yellow and blue. The information of the pH measurement was extracted from histograms of the pictures and a PLS model was performed to quantify the values. The results obtained were validated using reference methods, ion chromatography for phosphates and potentiometry for pH.
